چکیده

Osteosarcoma is the most common primary bone malignancy and primarily occurs in adolescents and young adults. Crude extract of Rheum palmatum L. (CERP) has been used as a traditional Chinese medicine for different diseases and there is experimental evidence that it may have anti-cancer effects. However, there is no information showing that CERP can affect the mobility of human osteosarcoma cells. In this study we determined the effects of CERP on U-2 OS human osteosarcoma cells. We found that CERP significantly inhibited the migration and invasion of U-2 OS cells. CERP also reduced the activity of matrix metalloproteinase (MMP)-2 and MMP-9 and decreased expression levels of the proteins FAK, GRP78, PKC, HIF-1, SOS1, VEGF, PI3K, GRB2, Ras, p-ERK1/2, ERK1/2, p-p38, JNK1/2, p-JNK1/2, MEKK3, MKK7, PERK, p-PERK, iNOS, COX-2, NF-kB p65, IRE-1, UPA, and RhoA in U-2 OS cells. Confocal laser microscopy revealed that CERP decreased the expression of NF-kB p65, RhoA and Rock 1. These in vitro studies suggest that CERP may gical Science and Technology, China Medical University, No 91, Hsueh-Shih Road, Taichung laureate Chinese Medicine, China Medical University, No 91, Hsueh-Shih Road, Taichung tw (K.-W. N-myc downstream regulated gene 2 overexpression reduces matrix metalloproteinase-2 and -9 activities and cell invasion of A549 lung cancer cell line in vitro

Objective(s):N-myc downstream regulated gene 2 (NDRG2) is a candidate gene for tumor suppression. The expression of NDRG2 is down-regulated in several tumors including lung cancer. The aim of this study was to explore the effect of NDRG2 overexpression on invasion, migration, and enzymatic activity of matrix metalloproteinase-2 (MMP-2) and -9 (MMP-9) in human lung adenocarcinoma A549 cells.  Ma...

Deguelin inhibits the migration and invasion of U-2 OS human osteosarcoma cells via the inhibition of matrix metalloproteinase-2/-9 in vitro.

Osteosarcoma is the most common malignant primary bone tumor in children and young adults and lung metastasis is the main cause of death in those patients. Deguelin, a naturally occurring rotenoid, is known to be an Akt inhibitor and to exhibit cytotoxic effects, including antiproliferative and anticarcinogenic activities, in several cancers. In the present study, we determined if deguelin woul...

Disruption of IGF-1R signaling by a novel quinazoline derivative, HMJ-30, inhibits invasiveness and reverses epithelial-mesenchymal transition in osteosarcoma U-2 OS cells

Osteosarcoma is the most common primary malignancy of the bone and is characterized by local invasion and distant metastasis. Over the past 20 years, long-term outcomes have reached a plateau even with aggressive therapy. Overexpression of insulin-like growth factor 1 receptor (IGF‑1R) is associated with tumor proliferation, invasion and migration in osteosarcoma. In the present study, our grou...

The Effects of NDRG2 Overexpression on Cell Proliferation and Invasiveness of SW48 Colorectal Cancer Cell Line

Background: Colorectal cancer (CRC) is one of the most common causes of cancer-related death in the world. The expression of N-myc downstream-regulated gene 2 (NDRG2) is down-regulated in CRC. The aim of this study was to investigate the effect of NDRG2 overexpression on cell proliferation and invasive potential of SW48 cells. Methods: SW48 cells were transfected with a plasmid overexpressing ND...

shRNA-mediated downregulation of α-N-Acetylgalactosaminidase inhibits migration and invasion of cancer cell lines

Objective(s): Extracellular matrix (ECM) is composed of many kinds of glycoproteins containing glycosaminoglycans (GAGs) moiety. The research was conducted based on the N-Acetylgalactosamine (GalNAc) degradation of ECM components by α-N-acetylgalactosaminidase (Nagalase) which facilitates migration and invasion of cancer cells. This study aims to investigate the effects of Naga-shRNA downregula...
